NMN Doubles Circulating NAD+ in Healthy Adults

65 participants, Avg age 34.714 days, 1,000 mg/day Randomised open-label RCTNMN supplementation doubled circulating NAD+ concentrations in healthy adults within 14 days, outperforming NAM (standard niacinamide) which showed no significant NAD+ increase.

Dose-Dependent Trial — NAD+ Levels & Health Scores

80 participants, ages 40–6560 days 300 / 600 / 900 mg/day. Multi-centre double-blind RCTNMN significantly raised blood NAD+ at all doses. The 900 mg group showed the strongest health score improvements — the 300 mg group improved walking speed by 24%, the 600 and 900 mg groups by 50%. Published in Geroscience.

Amateur Runners — Aerobic Capacity & Endurance

48 participants, ages 27–506 weeks 300 / 600 / 1200 mg/day. Double-blind RCTNMN significantly improved VO₂max and endurance in trained recreational runners. Higher doses produced greater improvements in oxygen utilisation. Published in the Journal of the International Society of Sports Nutrition.


Older Adults — Walking Speed & Sleep Quality

Ages 65–7512 weeks 250 mg/day. Randomised placebo-controlled double-blind. After 12 weeks, the NMN group had significantly faster 4-metre walking speed and improved sleep quality versus placebo. Blood NAD+ and its metabolites also rose significantly, confirming real bioavailability in older adults.



High-Dose Safety Study — 1,250 mg/day

31 participants, ages 20–654 weeks 1,250 mg/day Double-blind placebo-controlled RCTOral NMN at 1,250 mg/day did not result in any changes exceeding physiological norms across hematological, biochemical, urine, and body composition analyses. Confirmed safe even at high doses with no adverse effects.




Postmenopausal Women — Insulin Sensitivity & Muscle Health

25 participants, Prediabetic postmenopausal women, 10 weeks, 250 mg/day, Washington Univ. double-blind RCT. Skeletal muscle insulin sensitivity improved by approximately 25%. First study to demonstrate a real metabolic benefit of NMN in humans. NMN also upregulated genes involved in energy metabolism and muscle remodelling. Published in Science, 2021.

36 participants, Healthy middle-aged, 12 weeks, 250 mg twice daily. Double-blind placebo-controlled RCTNAD+-boosting supplementation with NMN showed significant improvements in well-being markers and blood NAD+ levels over 12 weeks in healthy middle-aged participants.

437 total patients, 10 RCTs included. Mean age 58 years. Mean follow-up 9.6 weeks. Dosage 150–1,200 mg/day PRISMA-guideline review · PubMed / NCBIAcross 437 patients, NMN consistently improved physical performance parameters, including muscle strength, endurance, and mobility. The safety profile was confirmed as excellent across all dose ranges (150–1,200 mg/day) with no severe adverse events reported in any included study.
